Sexual Health & Melanocortin Peptides

This class spans three distinct mechanistic families: melanocortin receptor agonists (PT-141, MT-I, MT-II), kisspeptin/KISS1R agonists, and GnRH receptor agonists. Two of five compounds are legitimately approved drugs — but for narrow indications that grey-market users routinely ignore. The melanocortin tanning peptides carry a plausible melanoma signal, and the HPG-axis peptides are frequency-dependent: the same molecule stimulates or suppresses depending on dosing pattern.

Compounds Covered (5)

PT-141 / Bremelanotide (Vyleesi)

Synthetic cyclic heptapeptide and non-selective melanocortin receptor agonist. FDA-approved for acquired, generalized HSDD in premenopausal women. Centrally acting — unlike PDE5 inhibitors.

FDA ApprovedPhase 3 Trial

Melanotan I / Afamelanotide (Scenesse)

Synthetic linear tridecapeptide, relatively MC1R-selective agonist. FDA-approved for erythropoietic protoporphyria (EPP), NOT for cosmetic tanning.

FDA ApprovedPhase 3 Trial

Melanotan II (MT-II / "The Barbie Drug")

Synthetic cyclic heptapeptide, NON-SELECTIVE melanocortin agonist. Not approved by ANY regulator. FDA classified as "unapproved new drug" — marketing is illegal. Owner criminally convicted 2015.

Not ApprovedPhase 1 Trial

Kisspeptin-10 (KISS1R agonist)

Decapeptide that acts as the "master switch" atop the HPG axis. FDA PCAC voted AGAINST adding to 503A bulks list (Oct 2024). Research-chemical only.

Not ApprovedPhase 2 Trial

Gonadorelin (Synthetic GnRH)

Synthetic decapeptide identical to endogenous GnRH. Its effect is ENTIRELY frequency-dependent: pulsatile = stimulation; continuous/too-frequent = suppression. No FDA-approved human product currently marketed.

Not ApprovedPhase 3 Trial

Comparison Notes

PT-141 (Vyleesi) and afamelanotide (Scenesse) are FDA-approved for specific narrow indications. Melanotan II is illegal to market and carries documented severe harms. Kisspeptin-10 was voted AGAINST for compounding. Gonadorelin has no marketed human product in the US.
